Overview
1-Hydroxybenzotriazole hydrate (CAS 80029-43-2 ), commonly sourced as HOBt hydrate, is a coupling additive used in peptide synthesis and pharmaceutical manufacturing. It suppresses racemization and activates carboxyl groups during amide bond formation.
This chemical directly impacts the purity and yield of active pharmaceutical ingredients. In solid-phase and solution-phase peptide synthesis, HOBt hydrate is used alongside carbodiimide reagents such as DCC or EDC to produce coupling-ready active esters.
These reactions result in reduced epimerization during the manufacturing process. API synthesis routes for small-molecule drugs rely on it to achieve high-purity amide intermediates under controlled conditions for various applications.
Contract manufacturers and research organizations use it in the preparation of peptide-based therapeutics. Stereochemical integrity at each coupling step is a critical quality parameter for these complex medical products.
It also supports the synthesis of peptidomimetics and specialty heterocyclic intermediates in medicinal chemistry programs. HOBt hydrate is supplied as a white to off-white crystalline powder for industrial use.
The hydrate form is the standard commercial offering, preferred over the anhydrous grade for its improved handling safety profile in bulk storage and transport. It is available in research quantities through to multi-kilogram lots.
Specifications are typically aligned to pharmaceutical-grade purity requirements, often reaching levels of ≥98% by HPLC. This ensures consistency and reliability for sensitive chemical synthesis and large-scale production cycles.
